The complement system is a complex series of reactions involving a cascading network of plasma proteins that help antibodies neutralize enemy antigens. During activation, Complement 5a (C5a) – one of the most potent pro-inflammatory mediators in the activation sequence – and other proteins interact with cells and initiate inflammatory and cellular immune responses to infectious organisms and injury.

Since excessive levels of C5a may be associated with autoimmune and chronic inflammatory disorders, inhibiting receptor activation may have therapeutic potential for such diseases as rheumatoid arthritis, systemic lupus erythematosus, reperfusion injury and acute respiratory distress syndrome.

The inventors developed potent, orally bioavailable C5a antagonists around which the University of Queensland formed Promics Pty. BioVentures Australia supported the company's work to test its lead compound and potential derivatives in animal models of inflammation.

In May 2006, Peptech Ltd. (ASX:PTD) acquired Promics. Peptech is continuing development of PMX53, Promics' most advanced drug candidate. PMX53 has successfully completed Phase I testing for safety and Peptech intends to advance the product to Phase II clinical trials for selected indications following the completion of additional formulation and dosing studies.
